12. Antigenic comparison of Akabane virus isolates by dot immunobinding assay

Neutralizing monoclonal antibodies (MAbs) against Akabane(AKA) virus were prepared. Competitive binding assay of these MAbs revealed that seven neutralizing epitopes exist on the G1 glycoprotein of the virus. Dot immunobinding assays (DIAs) using MAbs which recognized respective neutralizing epitopes were performed. Sixty-three isolates of AKA virus were divided into five groups according to their response patterns. The results show that DIAs can be used to effectively compare the antigenicities of Akabane virus isolates within a few hours, even with lesser amounts of virus culture than required for other assays.
