08. Experimental induction of AA amyloidosis by bovine amyloid fibrils in Sore Hock rabbits

 Experimental amyloidosis was reproduced by administration of bovine amyloid fibrils to rabbits afflicted by Sore Hock (SH), which is an ulcerative pododermatitis. Two groups of SH-afflicted rabbits were subjected to five inflammatory stimulations at intervals of 4 days by intraepithelial injection of a mixture consisting of Freund's complete adjuvant and lipopolysaccharide. One group of rabbits was administered amyloids in conjunction with the last inflammatory stimulation and the other group was not. Two groups were designed as additional controls : one (group 3) consisting of rabbits without SH that were subjected to five stimulations and administered amyloids, and another (group 4) consisting of SH-afflicted rabbits subjected to 0-4 stimulations and administered amyloids. Amyloid depositions were observed in all SH-afflicted rabbits that had been stimulated five times and given amyloids (18/18). In group 4, only one rabbit, which had been subjected to four stimulations, showed amyloid depositions. No amyloid depositions were observed in the other rabbits. In other experiments using pigs in place of rabbits, experimental amyloidosis was not reproduced. There might be some species differences in the pathogenesis of AA amyloidosis.
