01. Development of a genotyping method for predicting the serotypes of Streptococcus suis

We developed a practical and easy two-step multiplex PCR assay to help serotyping of Streptococcus suis. First, PCR detects the capsular polysaccharide synthesis (cps) genes conserved in multiple serotypes and classifies the tested strains into seven groups (grouping-PCR). Second, PCR detects the cps genes specific to respective serotypes in each group and identifies the cps-type of the strain (typing-PCR). The assay accurately typed almost all of the serotype reference strains and field isolates of various serotypes (355/371; 95.7%) and can identify the genotypes of cps gene clusters of some serologically non-typeable strains. Our data strongly suggest that the developed cps typing methods are practical and can further contribute to the diagnosis and epidemiological studies of S. suis infections.
