Acute phase responses in cattle through the course of splenectomy and Theileria orientalis infection (20分)

Induction of acute phase responses was investigated in cattle through the experimental course of splenectomy and infection with Theileria orientalis. Four and one Holstein calves were splenectomized by surgical operation, then inoculated with 109 and 1011 parasitized red blood cells (RBC) 7-8 weeks after operation, respectively. Blood samples were collected 1-3 times a week to determine RBC and white blood cell (WBC) counts, hematocrit (HCT) value, hemoglobin (HGB) concentration, mean corpus volume of RBC (MCV) and population rate of lymphocytes/granulocytes (%LY/GR) using an automatic hemocytometer, serum concentrations of α1 acid glycoprotein (α1AG) and haptoglobin (Hp) by gel immunodiffusion method. Parasitemia was evaluated by observation of blood films. Parasites were first detected in blood films 12-14 and 3 days, and anemia showing an HCT value less than 25% was developed from 4-6 weeks and 10 days after infection in animals given a low dose and high dose of parasites, respectively. WBC counts did not change through the experiment except for a decrease detected 3 weeks after infection. %LY/GR became inverted transiently between 1 and 3 days after splenectomy, showing a decrease in %LY and an increase in %GR. Thereafter, %LY increased and %GR decreased 10 weeks after infection. A slight and transient rise in α1AG was detected 3 days after splenectomy. α1AG levels increased in the late stage of infection, from 8 weeks in animals given a low dose of parasites and from 3 weeks in an animal given a high dose. A remarkable but transient rise in Hp was detected between 3 and 7 days after splenectomy, and the levels returned to an initial undetectable level. Hp appeared again only in one animal 8 weeks after infection. In the course of infection, α1AG levels were correlated with RBC counts, HCT and MCV values, HGB concentrations and %parasitemia. These results suggested that different types of inflammation are induced in the course of splenectomy and T. orientalis infection, and the increase in α1AG with no Hp induction in T. orientalis infection is related to parasite growth and the level of anemia.
