▼ RNA依存性DNAメチル化によるエピジェネティック転写活性化機構の解明
当研究室では、RNA依存性DNAメチル化(RdDM)を介する分子機構によって、ペチュニアの花のホメオティック遺伝子pMADS3の転写が活性化する現象を見出しました。RdDMが遺伝子サイレンシングに関与することは知られていましたが、遺伝子サイレンシングとは逆に、転写の活性化を誘導したという現象は前例がありません。pMADS3の第2イントロン中の1カ所のシトシンのメチル化が原因であるところまではつき止めましたが、それがなぜ転写活性化に至るかという部分は未解明です。このような制御が生物学的にどのような意味をもつのかを解明したいと考えています。また、この転写制御はDNAメチル化によってゲノム上に記憶され、組換え遺伝子の分離後も形質が残るため、非遺伝子組換えによる植物の形質改良法に発展する可能性
